چکیده
Numerous patients experience articular cartilage defects (ACDs), which are characterized by progressive degradation and often lead to osteoarthritis (OA). Consequently, 44.7% of OA suffer from dyskinesia or disability. Current clinical drug treatments offer limited effectiveness in fully curing the disease. In this study, we propose a collaborative approach that combines physical biological cues promote regeneration. A biodegradable piezoelectric poly (l-lactic acid) (PLLA) nanofiber scaffold facilitates situ, battery-free electrical stimulation under natural joint loading, while extracellular vesicles (EVs) serve as communication mediators between cells cell proliferation, migration, secretion type II collagen. combined approach, EVs attached PLLA gradually released localized taken up chondrocytes. This process results organization collagen along fiber surface, ultimately forming lacunae facilitate residence new As an outcome, significant round defect (diameter: 3 mm depth: 1 mm) PLLA/EVs group (rat knee) was rapidly restored within six weeks. contrast, individual groups demonstrated considerably weaker regeneration capabilities. research suggests synergistic effect electromechanical EVs-based is crucial intervention method for treating OA.

Mesenchymal Stromal/stem Cell-derived Extracellular Vesicles Promote Human Cartilage Regeneration In Vitro
Osteoarthritis (OA) is a rheumatic disease leading to chronic pain and disability with no effective treatment available. Recently, allogeneic human mesenchymal stromal/stem cells (MSC) entered clinical trials as a novel therapy for OA. Increasing evidence suggests that therapeutic efficacy of MSC depends on paracrine signalling. Here we investigated the role of extracellular vesicles (EVs) secr...

The fact that bone is stress electrogenic is well substantiated. Becker and colleagues have proposed that this property is the biological signal that regulates Wolff's law, whereas others have observed osteogenic responses to electrical voltages and currents. Becker and Murray detected electrical control signal characteristics in fractured long bones. The above findings and the work of Smith, w...
متن کاملEXTRACELLULAR VESICLES AND THEIR THERAPEUTIC APPLICATIONS: A REVIEW
Background & Aims: Extracellular vesicles are double phospholipid layer vesicles releasing from several cells. These particles contain bio-molecules including mRNA, miRNA, DNA, proteins, and lipids which play a pivotal role in the cell to cell communication. Once delivered, they participate to change target cell function, fate, morphology, differentiation, and growth. The present study is a re...
متن کاملStem Cell Extracellular Vesicles: Extended Messages of Regeneration
Stem cells are critical to maintaining steady-state organ homeostasis and regenerating injured tissues. Recent intriguing reports implicate extracellular vesicles (EVs) as carriers for the distribution of morphogens and growth and differentiation factors from tissue parenchymal cells to stem cells, and conversely, stem cell-derived EVs carrying certain proteins and nucleic acids can support hea...
متن کاملIsolation and characterization of Faecalibacterium prausnitzii extracellular vesicles
Introduction: Extracellular vesicles (EVs) contain active biological compounds which play important roles in biological processes. The secretion of EVs is a common phenomenon which occurs in archaea, bacteria and mammalian cells. The secretion of bacterial EVs has been discovered in various species of both Gram-negative and Gram-positive bacteria. Faecalibacterium prausnitzii is one of the comm...
